Abstract

This is a sub-study in a project where the overall purpose is to study how mitochondrial energy metabolism is utilized in cells to produce one-carbon metabolites, how it is dissipated through the actions of UCPs, if it is associated to risk of diabetes, the metabolic syndrome, or cancer. We shall begin with analysis of existing genome-wide DNA methylation chip data of our own, run at SciLife Lab to study the methylation pattern of Uncoupling Proteins 1-5 (UCPs). Then, with the aid of publicly deposited data from tissues, cell lines and tumours we hope to identify Tissue-specific Differentially Methylated Regions (TDMRs) in the UCPs. These TDMRs will then by validated in our lab using sequence-specific quantitative DNA methylation assays by Pyrosequencing.
